To further extend the depth of coverage of the plasma proteome beyond the low ng/mL protein concentration limit obtained with the 1D standard-flow platform, we have developed a 2-D LC peptide fractionation method. The approach specifically involves 2 dimensions of chromatographic separation, with the first conducted at alkaline conditions and the second at acidic, prior to dynamic MRM/MS detection. This strategy has enable improved peptide selectivity for increased detection sensitivity down to the mid pg/mL level in undepleted and non-enriched human plasma.
